Mechanism of Action
Tropisetron Hydrochloride acts as a selective, competitive antagonist of the 5-hydroxytryptamine type 3 (5-HT3) receptor (IC50 = 70.1 nM), blocking serotonin action both peripherally on vagus nerve terminals in the gastrointestinal tract and centrally in the chemoreceptor trigger zone (CTZ) of the area postrema, the emetic center of the brain. It also functions as an α7-nicotinic acetylcholine receptor (α7-nAChR) agonist and calmodulin inhibitor, exerting neuroprotective and immunomodulatory effects. Unlike some antiemetics, it has minimal affinity for other receptors (5-HT1, 5-HT2, dopamine, histamine), resulting in fewer extrapyramidal side effects. It is mainly metabolized by cytochrome P450 (CYP) 2D6 in the human body, with a half-life of approximately 8-10 hours.
